Explain the principle of haplo-insufficiency profiling (HIP) in yeast
Question Description
Genomics course
Aims and Objectives; In the lectures we discussed the use of bar-coded gene deletions in yeast to monitor the relative fitness of 4500 deletant strains, in competitive co-culture. Such massively parallel phenotypic profiling can be used in chemogenomic screens, to identify which proteins are targeted by a given drug using the HIP and HOP processes. Here we will discuss the results of an investigation of the sensitivity of yeast to a range of psychoactive drugs, including Prozac, using HIP and HOP screens. The screen aims to identify the non-intentional, ‘off-pathway’ drug protein interactions.
The Question: Explain the principle of haplo-insufficiency profiling (HIP) in yeast (guidance: <200 words).
